The Janus kinase/signal transducers and activators of transcription (JAK/STAT) pathway is a chain of interactions between proteins in a cell. This signaling pathway transmits information from extracellular chemical signals to the nucleus as a result in DNA transcription and expression of genes involved in proliferation, differentiation, immunity, apoptosis and oncogenesis. There are four JAKs (JAK1, JAK2, JAK3, TYK2) and seven STATs (STAT1, STAT2, STAT3, STAT4, STAT5a, STAT5b, STAT6) in mammals. This pathway plays an important role in transduction for cytokines. JAK phosphorylation of the STAT proteins then makes dimer complex, and translocates to the nucleus. Once in the nucleus, STAT dimmers are stabilised by NT:NT interactions and bind to tandem sequence elements within promoter regions and activate the transcription of specific gene subsets. Dysregulation of JAK-STAT pathway leads to various disorders like rheumatoid arthritis, systemic lupus erythematosus, Crohn’s disease and Sjogren’s syndrome, and STAT6 SNPs is associated with asthma. Many drug companies have started to develop therapeutics drug which inhibit the function of JAK tyrosine kinases.
